Tubulin (Tub) is one of the most abundant proteins in eukaryotic cells and a major target of antimitotic drugs. Tub is a dimer of α- and β-tubulin and heterodimers have been isolated from cell and crystallographic studies. The conformation and dynamics of the tubulin dimer and the interactions between tubulin subunits are of importance. We have used steady-state, stopped flow and single-molecule fluorescence spectroscopy to study the interactions between tubulin subunits and dimers. Tubulin assembly results in a loss of fluorescence from the fluorophores on both α- and β-tubulin, as well as a concomitant decrease in the blue-shifted photoluminescence. Tubulin subunits, tubulin dimers, and combinations of each have been studied. We find that in the case of TubA – TubB, there is no quenching or blue-shifted emission. For dimers of α- and β-tubulin tubulin we observe quenching of the alpha photoluminescence upon tubulin polymerization. The populations of unquenched and quenched dimers depends on the overall kinetics of the experiment, supporting our hypothesis that the quenching is due to photochemical quenching of the fluorophores in the core of the dimer. When the dimer is quenched the photoluminescence of the beta fluorophore is increased and that of the beta fluorophore is decreased. Since tubulin has been shown to “autocatalytically” dimerize we conclude that quenching is due to inter-tubulin dynamics within the dimer. We observe that the combination of β-tubulin with a heterodimeric tubulin shifts the tubulin peak to a shorter wavelength
